To evaluate the pharmacokinetics of SFX in the blood, the bioavailability of sildenafil citrate was studied in the presence of a positive and a negative dose of sildenafil. In vitro bioavailability studies were conducted in normal and in patients with normal serum sildenafil concentrations (100 and 250 ng/ml, respectively) (SFX). In vitro kinetics was assessed by comparing the terminal half-life of sildenafil in solution with that in the plasma. The plasma half-life of sildenafil citrate was approximately 6 hours and its concentration was steady in healthy volunteers.
In vitro pharmacokinetic studies, in patients with normal and in patients with normal serum sildenafil concentrations (250 ng/ml, i.e. 100 ng/ml) (SFX) and in patients with a history of erectile dysfunction (ED, i.e. a positive dose) showed that a single dose of sildenafil citrate produced a mean peak plasma concentration (Cmax) of 4.5 ng/ml and a mean plasma concentration of 8.0 ng/ml in ED patients and healthy volunteers, respectively. The Cmax in healthy volunteers was not different from that in ED patients. In patients with a history of ED, the mean peak plasma sildenafil concentration was 1.8 ng/ml and the mean Cmax was 5.8 ng/ml.
The pharmacokinetics of sildenafil citrate were evaluated in healthy volunteers (n=14) and in patients with ED (n=14), using sildenafil citrate as a positive and a negative dose of sildenafil. The plasma half-life of sildenafil was approximately 6 hours in healthy volunteers (median 2.5 hours) and was steady in healthy volunteers.
SFX is an active, highly active and well absorbed drug. It was shown to have a good pharmacokinetic effect in healthy volunteers and to have a good pharmacokinetic effect in patients with a history of ED. Therefore, the pharmacokinetic disposition of sildenafil in healthy volunteers was not different from that in patients with ED. The pharmacokinetics of sildenafil citrate in the blood were evaluated in patients with normal and in patients with normal serum sildenafil concentrations.
